FUSIONPlex™ Heme v3 & Pan-Heme v2

RNA-based next-generation sequencing solution for the analytically sensitive identification of known and novel gene fusions, splicing variants, exon-skipping events, SNVs/Indels and gene expression in haematological malignancy research.

FUSIONPlex Pan-Heme v2 provides broad coverage of 218 genes associated with haematological malignancies, while FUSIONPlex Heme v3 provides more focused coverage of 121 genes targeting alterations frequently found in leukaemia while retaining markers relevant to other haematological diseases.

FUSIONPlex Heme v3 and Pan-Heme v2

Detailed Description

Operating Principle

The Archer FUSIONPlex Heme v3 & Pan-Heme v2 solutions are based on Anchored Multiplex PCR (AMP™) chemistry, a proprietary multiplex PCR strategy that uses unidirectional gene-specific primers to enrich target regions and enable the detection of known and unknown alterations. The adapters incorporate molecular barcodes (MBCs, Molecular Barcodes) and sample indices, enabling multiplexed analysis, read deduplication, error correction and high-confidence variant calling.

The approach is designed for RNA fusion analysis, enabling expressed fusion transcripts to be captured and avoiding the technical and economic difficulties associated with sequencing large repetitive intronic regions in DNA-based assays. In combination with de novo assembly and Archer Analysis software, FUSIONPlex enables the sensitive identification of known and novel fusions, including fusions with previously unidentified partners (fusion-agnostic).

Benefits

Enables multiple types of alterations to be characterised in a single workflow, reducing the need for separate assays.
Identifies known and novel fusions, including complex fusions or those with variable partners.
Supports panel content customisation to incorporate new biomarkers or adapt to specific areas of research.
Compatible with manual or automated library preparation; consult the available automation options.
Fast and simple manual workflow: 1.5-day library preparation with 3.5 hours of hands-on time. Uses colour coding and lyophilised reagents for a simple and intuitive workflow.
Integration with Archer Analysis, a secondary analysis platform designed to simplify interpretation and generate high-level and detailed mutation reports.

Technology Used

RNA-based next-generation sequencing (NGS). Anchored Multiplex PCR (AMP™) chemistry. Gene-specific primers (GSPs) for targeted enrichment. MBC adapters for molecular tagging, read deduplication and error correction. Archer Analysis for bioinformatics analysis, visualisation and interpretation support. FUSIONPlex panels compatible with libraries generated from RNA or total nucleic acid (TNA), depending on the protocol and application.
