Comprehensive system for NGS methylation analysis combining enzymatic conversion, target enrichment, and high-performance methylation panels to identify methylated regions with greater efficiency, less DNA damage, and optimized sequencing metrics. The solution is designed for epigenetics, oncology, and the discovery of differentially methylated regions.
Detailed Description
Operating Principle
This is an end-to-end sample preparation and enrichment solution geared towards methylation analysis.
The workflow combines an enzymatic conversion of unmethylated cytosines, followed by capture-based enrichment using Twist probes and optimized hybridization reagents. Compared to chemical bisulfite treatment, this enzymatic approach is less harsh on DNA, requires less sample input, and generates higher quality libraries with better yield. Following PCR, the nucleotide representation is equivalent to that of bisulfite-treated DNA, allowing it to be integrated into established bioinformatics pipelines.
Clinical Applications
Benefits
The system stands out for providing a less damaging library preparation, with better mapping efficiency, higher GC uniformity, and overall improved sequencing metrics. In addition, it incorporates the Twist Methylation Enhancer, a specific blocker for post-conversion samples that reduces off-target capture by up to half without significantly affecting other metrics.
