NGS Shotgun sequencing is a laboratory technique for determining one or more DNA sequences from the genome of an organism or an entire sample. The method consists of breaking the genome into a collection of small DNA fragments that are arranged individually. The individual fragments must then be bioinformatically placed in the correct order to reconstruct the genome.

Illumina offers reagents to prepare libraries from any sample without prior culture.

Depending on the target/information to be obtained, the number of reads/sample will vary. It will also depend on the size of the genome. Typically, ShotGun studies start from 100 million reads/sample; therefore, large sequencing platforms (NextSeq/NovaSeq) are usually required.
